Very little is known about the history of Alexei Luthor before he rises to power to take control of several minor European nations prior to World War 2 through his super-scientific mechanitions. He intended to plunge all of the Europe countries into war starting with the small countries of Galonia and Toran. He used his scientific genius to sabotage a peace conference but was defeated by Kal-L the Superman native to his Earth 2. Alexei escaped and later plagued Kal-L with a gigantic Kryptonite meteor after learning about the Earth 2 Superman's weakness from Dan Rivers. Not able to defeat Superman, Alexei took solace in publically revealing Superman's weakness the general world which caused trouble to Kal-L from various other foes.
Alexei would returne time and time again in elaborate plans to kill Superman but was defeated each time though he usually achieved better than outright defeat.
Later in his career, Alexei would learn of the multiverse and teamed up with his Earth 1 counterpart in an attempt to conquer Earth 3. Recruiting Earth 3's primary villian Ultraman (villianous counterpart to their Superman) to aid them in conquering Earth 3, Alexei and Lex Luthor were defeated by their Earth 3 counterpart, Alexander Luthor, Sr., who became Earth 3's first and only superhero.
Alexei would continue on his private quest for world domination of his home Earth 2 until he was recruited by Brainiac during "The Crisis of Infinite Earths". Upon confronting Lex Luthor who had been selected over him by Brainiac as field commander of their amassed gathering, Alexei stated to all present "I'm better than that bloody labrat! You don't need two Luthors!" to which Brainiac agreed with, and immediately killed Alexei Luthor leaving Lex the only Luthor present.
